Does the bulb flash if the 4-way emergency flasher lights are operating? With the turn signal flashing?
You need to use a voltmeter to test the contact for the bulb to the ground side and see if there's voltage. Then if not, check to be sure there's voltage to a known good metal ground on the body.
I've seen connectors in the lines for parking lights and turn signals on the front that are exposed to the elements that have connection problems within the connector that's close to the light housing. But brakes are usually inside the vehicle in the trunk. So go to the connector lines if you're not getting appropriate readings on the bulb contacts when the brake light should be on or turn signal is on if it's the same bulb.
I'm not clear what you mean by signal bulb holder. Are the brake bulb and signal bulb the same in this case?
