transmission - '06 Sequoia

I have a 2006 2wd Sequoia limited. Any time the transmition is engaged there is a major vibration. If I knock it into neutral while it is rolling the vibration goes away. The vibration is present if the transmition is pulling and when it is downshifting. Anytime the is a load of any kind. Truck has about 260,000 miles on it. Does anyone know what is causing this problem and what I need to do to correct it?
